Protein stability
The advantage of doing formulation screening in C3 is that we use our HTP robotics and combinatorial software to keep the assays fast and reproducible. Our formulation screening is based on thermal stability, and can help you define a suitable base-formulation that will keep a protein stable and at high concentrations for extended periods, making it more useful (and reproducible) for crystallisation trials, binding assays and activity assays. This assay is accessed using the intuitive booking site, and the data can be interpreted via software that is easily downloaded from us.
Formulation Screening
The method is based on Differential Scanning Fluorimetry (DSF; a.k.a. Thermofluor), with our standard screen using 14 buffers over a range of pH at high and low salt concentrations. We’ve worked hard to make the method robust, so that you can shift to a novel formulation with confidence.
We’ve selected the formulations to be specifically kind to protein purification methods and subsequent crystallisation trials, look for ‘Buffer Screen 9’ under available screens on the C6 webtool.

Optimisation, Additives & Ligands
Once a good base condition has been found, we can use the same technology to look at how known co-factors or any number of possible additives and ligands (found in the C3 additive screens, for example) enhance thermal stability of your protein.
Alternatively, we can use the screen optimisation tools available for protein crystallisation to create bespoke buffer screens.
